Drug of choice is basically someone’s favorite recreational drug, and first line medicine is what is preferred clinically to be used before trying other drugs.

I like your definition of "drug of choice", i thought of it as the doctors "drug of choice".
 
Drug of choice for a disease doesn't usually work, as it depends on the current situation I would say. lets say first line didn't work, what's the drug of choice then?

So forget about "drug of choice", its a bad term. unless you use it like @Singh
